Purpose
It is a UAS reporter construct (5×UAS) that expresses a GFP nanobody fused to a 3×NLS sequence with a 6D linker. The pUASt-attB (5xUAS) backbone was a gift from Konrad Basler (Bischof et al., 2007) and we have inserted the kozak_3xNLS-6D-GFPnanobody sequence. It accelerates nuclear labeling in embryos by eliminating the lag associated with GFP maturation (when coupled with maternally deposited GFP). It converts cytoplasmic GFP expressed in transgenic fly lines into a nuclear label.
