Notes for the Kaufman Lab
There are 2 glycerol stocks of this vector: Clone 1 and Clone 5. The vector was first published in the Holtzman et al, 2010 Fly paper and actually described in full in the Kuwayama et al, 2014 PLOS One paper. There is no sequence or map data except what Niimi provides in the 2014 paper. One could theoretically reconstruct the vector from the M&M cloning section. Wimmer may have posted sequence data for his original vectors used as source material. In the Holtzman paper, we used the vector to generate lines expressing the transposase, which is marked by CFP. This part of the vector was referred to as: pBac{5pBlueEye} = pBac(hsp70-transposase)::(3xP3-ECFP). This is half of the vector was referred to as: (original construct = pBacâ¢(hsp70-transposase)::(3xP3-ECFP)::pBacâ¢(hsp27-EGFP)::(DsRed-3xP3)pBac). Niimi's nomenclature in 2014 is slightly different: pBac(hsp70-transposase)::(3xP3-ECFP)::hsp27-EGFP::(3xP3-DsRed). These are the same vector but the different nomenclature has the potential to cause confusion.
