For creating the pUASp-AttB vector, we amplified a 375- bp fragment containing the attB sequence, which was added with AatII sites at both ends and subcloned into the AatII site of the pUASp vector.
Note that you are no longer able to use the Kpn1 site in the MCS as there is a Kpn1 site within the attB.
